I think my second most satisfying moment was when I got my first pokemon to level 100.

Shooting Handsome Jack in the face while he was mid sentence. It took over a year as it was a co-op playthrough with my partner and she tended to lose interest after a while but when we finally got to him... Oh it felt good.
