What does it mean in more depth to have 'one up' on someone when it comes to attracting the opposite sex.

It means you have more positive qualities than the other person which would attract the opposite sex.

You might have a better body than the next guy.

Or have a better mind.

Or just be a nicer guy.

Or have more money.

Or be better educated.

Or live in a nicer house (sans parents).

Or drive a nicer car.

To "have one up" on someone else is to have an advantage over them.

To "one up" someone is to perform better than they.

One-upmanship is the art or practice of successively outdoing competitors.
