I get what he's saying, but it makes him sound not very manly. :D Dudes don't say things like that. It's kinda fussy and overly dramatic and unnatural coming from a man. Of course this is a joke so it's made up.

It isn't funny on its own but when preceded by a setup that makes it humorously obvious why she's leaving him (and it isn't for another man) then it's funny.

An example I've seen (but couldn't find a google image for) is she walks in on him having sex with another woman and he says "who is he?" while literally in the midst of sex with this other woman.
