If you were in a very heated argument with a guy that you were in a relationship with and the guy said:

" you think that you're a man but you need to realize you aren't!"

What would you're reaction be?

Confusion. There are so many things that statement could mean.

Something along the lines of "What on earth does that have to do with anything that we're discussing?"

To break up with them because they sound like a misogynistic jerk?

Or maybe not depending on the context, like if that was in response to me thinking I could impregnate a woman with my nonexistent sperm. Then what they said would make sense.

I can't say for sure without knowing what it was about.

To me it almost sounds like a straight guy having an argument with a lesbian who wants to have a baby with another woman.

QFT.
My reaction would be to ask him what his definition of a man was, and how I was infringing on that in his mind.

I don't really understand what that means. I'm guessing because she's taking the "roles" that men are supposed to fit or something. Maybe because he believes in traditional gender roles?

I would say "If you think gender has anything to do with this fight then you are a larger idiot than I presumed. If you were truly the man you claim to be then you would be able to support a compromise we would make together." The context is vague but it sounds nice.

In most contexts, the statement would be misogynistic.
I suppose there are a few instances in which it would be less offensive, such as exhorting a woman to stop trying to lift heavy things because she keeps hurting herself. Or maybe asking her to stop jogging alone at night through bad neighborhoods. In these contexts the statement expresses concern for her safety and well-being, though to be honest it still comes across rather poorly.

Laughing. Because your argument would mean, that you would think of certain reactions, oppinions or feelings being male and certain reactions being female. Elseway telling someone that her reactions gives you the impression, that she might think to be a male makes no sense.

One of the few things that would fully allow you to suggest, that an woman may wrongly think to be a man, is if she tried to open her zip and pee in an pissoire. ^^ Or other stuff involving direct action with certain genitals.

While if you try to negate someones oppinion, with the argument, that as a man/woman you are not allowed to have this or that oppinion or feelings, its simply nonsense. Maybe try to negate someones thoughts/oppinion/feelings with arguments next time. As long that in your world, that isnt part of strict female behavior and so would force you afterward to realize, that you are no woman, and are out of this reason not allowed to have the oppinion of using logic arguments.

It's hard to say without the context, but it seems that the guy is suggesting that she is behaving in an inappropriate way. I need more information.

It can hardly be about inappropriate behavior, because I would not know how an certain behavior, is either appropriate or inappropriate, only because of the location of some dangling genitals. ^^ By saying so: Nope, only because of male genitals being more easy to reach, its still not appropriate to pee in urban areas outside of an toilette. ^^
