In more depth what does it mean if somebody is "not your biggest fan"?

Erm I think it means that the person doesn't exactly like you.. That is what I think it means, not sure how else to explain that.

Hm oh that's interesting.. Maybe your uncle means it in a different way to how I think it means

Someone who isn't the biggest fan of something might be anything from totally against that thing to an average fan of it. Without further context or information on the relationships that you hold with your brother and uncle, it can only be said that your brother believes that, although your uncle may or may not like you, your uncle perhaps sees something in you which irks him or your uncle is simply not enthusiastic about spending time with you and learning about you. There's still a chance that your brother misinterpreted something that your uncle said though.

Ive heard this use in a similar situation, the child was being friendly to some one who was not receptive to him. It was being said to tell the child to back off or to avoid being to personal with him to avoid friction between them.

Its like "I could live without such-and-such".

Though "not your biggest fan" could mean that they are just an average fan of you ( as opposed to being one of your screaming groupies)- it rarely means that.

It usually means that they arent a fan of you AT ALL.
It could mean that they find you mildly irritating. Or it could mean that they hate your guts- or it could mean some shade and gradation in between.

But its a polite understated way of saying that you do something to rub them the wrong way to some degree somehow.
