A Genetic Clue to Why Autism Affects Boys More

Among the many mysteries that befuddle autism researchers: why the disorder affects boys four times more often than girls. But in new findings reported online today by the journal Molecular Psychiatry, researchers say they have found a genetic clue that may help explain the disparity.
The newly discovered autism-risk gene, identified by authors as CACNA1G, is more common in boys than in girls (why that's so is still not clear), and the authors suggest it plays a role in boys' increased risk of the developmental disorder. CACNA1G, which sits on chromosome 17, amid other genes that have been previously linked to autism, is responsible for regulating the flow of calcium into and out of cells. Nerve cells in the brain rely on calcium to become activated, and research suggests that imbalances in the mineral can result in the overstimulation of neural connections and create developmental problems, such as autism and even epilepsy, which is also a common feature of autism. "Our current theories about autism suggest that the disorder is related to overexcitability at nerve endings," says Geri Dawson, chief science officer of Autism Speaks, an advocacy group that provided the genetic data used by the study's authors. "It's interesting to see that the gene they identified appears to modulate excitability of neurons."
For the new study, researchers at the University of California, Los Angeles (UCLA), combed the genetic database of the Autism Genetic Resource Exchange (AGRE), a resource of DNA from 2,000 families with at least one autistic child. The scientists focused on the more than 1,000 genetic samples of families in which at least one son was affected by the disorder, prompted by the results of an earlier study using the same database, which identified a rich autism-related genetic region on chromosome 17 that contained genetic variants more common in boys than in girls. While nearly 40% of the general population has the most common form of CACNA1G, one variant of the gene was more prevalent in autistic boys, researchers found. "There is a strong genetic signal in this region," says Dr. Daniel Geschwind, director of UCLA's Center for Autism Research and Treatment and one of the study's co-authors. "But this gene doesn't explain all of that signal or even half of it. What that means is that there are many more genes in this region contributing to autism."
That's not surprising for a disorder as complex as autism — actually, a spectrum of developmental disorders involving impairment in language, social behavior and certain physical behaviors — with symptoms that range widely in number and severity. So far, studies have linked a handful of genes, all of which play a role in the way nerve cells connect and communicate, with autism spectrum disorders. It's likely not only that a large number of genes contribute to the disorder, but also that a different combination of genes — as well as unique interactions between genes and environment — are responsible for each individual case of autism.
So it's certainly a daunting challenge to begin teasing out the individual genes that may contribute to autism, as the UCLA team has with CACNA1G, but databases like AGRE make the job slightly easier. The next step will be to try to use known autism genes to help develop screening tools or early interventions. "We are going to have a much better understanding of the causes of autism over the next five to 10 years," says Dawson. "We're in a period of great discovery."

I don't mind research that leads to more early interventions, but not too fussed if they use this kind of info for the development of screening tools that might eventually render interesting /eccentric ASD people obsolete.

I'm trying to figure out how this could possibly work, how a gene on chromosome 17 could be more common in males. I can only think of two possible explanations.
It might happen at meiosis, during the formation of the egg or sperm cells. I don't see how the egg cells could be involved, since they all carry an X chromosome, gender is irrelevant in egg cells.
I guess it could be that when sperm cells are formed, the a copy of chromosome 17 that's carrying the CACNA1G gene is more likely to go along with the Y chromosome batch, but I'm not aware of anything along those lines normally happening.
Other people know more about this process than me, so maybe this is something that's known to happen, but I'm not aware of it.
The only other possibility I can think of is that for some reason, female embryos carrying the CACNA1G gene are much more likely to die at some point between conception and birth. I guess that's possible, but I don't see any reason why it would be likely.
I have a hard time making sense of this.
Also, this is just an impression of mine and I'm not claiming it's anything else, but it seems more likely to me that Asperger's/Autism sorts of conditions would be more likely to involve the structure of connections between brain cells rather than simply the rate of firing of individual cells, which is all the flow of calcium into and out of the cells would influence.

we can all be forgiven for misinterpreting: the suggestion would always be that it is about WHY autism would be more common in boys, which indeed would be what the average reader would expect from a NEWSpaper
but please read again: the article only report that there is, once more, another CLUE
well... yes indeed, ANY new genetic information would involve more knowledge about autism generally, and thus about this gender-dichotomy - the header should just have read: 'new genetic information on autism' - which is not really news, these days, or is it

Autism has exploded in the USA. So, either the autism gene is being spread at an epidemic rate (not likely) or environmental changes are triggering more autism cases (very likely). Girls have a "blood barrier" factor that filters out more contaminants and lets them endure certain toxins better than boys do. If autism is being triggered by an environmental factor, this explains why so many more boys than girls develop autism.
We're literally poisoning the population. We've got so many hormones in the drinking water (as waste treatment plants don't remove them in the filtration process) that it is considered a major cause behind the growing number of infertility issues couples suffer from.

The idea that autism is an epidemic is highly questionable. The best studies out there indicate that the rate really hasn't deviated much from 1 in 150 (give or take a few in either direction). Media, however, loves a story, so they have a tendency to pounce on information that appears sensational. My apologies to my friends in the media, but you know just as well as I do that it's true.

I can tell you that this kind of "epidemies" as you call them can be comparable in number of people that has it (diagnosed and undiagnosed together) from country to country in the whole world. The genetic heritage has a big rol on spreading it in the population so for example in my country Colombia ADHD prevalence in kids from a particular region of the country is 14% compared to 5% in another part of the country.
This happened thanks to cultural definitions of success. In this region where prevalence is 14% people's culture is about negociating, working energeticly non-stop, partying energetically non-stop, and taking risks; there having ADHD is seen as a good quality, even a charm.
In the part where they have 5% of prevalence what is considered socially accepted is studying, graduating, having everything under control, socially interating under control, being a more focused person, evaluating posibilities, getting a regular job and settling down. In this region ADHD people has less chance to spread their genes therefore they have less prevalence.
I'm talking about ADHD, but let's talk about AS. I think it's more socially accepted and even cooler to be aspie if you live in silicon valley than if you live in miami. In silicon valley your work results (or even your geekyness) can be determinating in your social acceptation or success. I'm naming silicon valley, but now change that name for any other state in USA where hard working or hard thinking is a good quality and there you got! This people is the people that has more chances to perpetuate their DNA (that of course even more after they have earn enough money to play Hugh Hefner
What I do agree is that diet can be determining as well. I can realize that omega3 and vitamin b complex helps me a little bit for ADHD as well as food rich in Phosphatidylserine and other complex vitamin acids and phospholipids that have been related to it. Never as good as ADHD medication though, good enough to recognize and accept that my brain feel better when I feed him better. None the less this dietary special requirements only improve it, do not cause it.
I close my post with the blue people of kentucky, people whose skin is blue tinted thanks to a recesive gene. They used to be seen as normal in a region where no one never came so they started breeding until their pool of genes came out of the isolation and so did the people, then they started to become fewer and fewer. their genes are not that popular any more :S

As regards the autism "epidemic", I feel I must once again throw a sense of historical perspective into the mix. Every study I've seen thus far regarding the "epidemic" compares diagnosis rates across decades - often 1980 vs 2005. However, these numbers did not occur in a vacuum, or under a state of constancy. The most important factor, I believe, would be the publication of new editions of both the ICD and the DSM, with new diagnostic criteria for autism (and including AS, NVLD, and PDD-NOS for the first time ever), during the early 1990s. Many patients who would have been dismissed as "eccentric" or misdiagnosed as "schizoid" <raises hand meekly> are now found, under the new criteria, to be autistic, or at least on the spectrum. Remember also that it took time for the new documents to filter their way into the psychiatric community - psychiatrists and psychologists are human too, and often unwilling to change their paradigms too quickly.
Similarly, one might compare the rates of diagnoses of mental illness between, say, 1950 and 1975, note the sharp drop, and ignore the fact that the DSM-II defined homosexuality as a mental illness, a classification that was removed in the DSM-III. This could lead to all manner of incorrect conclusions...
